I marathoned Working!! and Bakemonogatari last week.

Working!! was fun and chill to watch. Inami was the best girl and hopefully the protag will give her the dick and they'll live happily ever after. Yamada was also cool in a little sister you'd like to spoil with ice cream kind of way. Both seasons get 8 autism bucks out of 10 from me.

Bakemonogatari was weird as fuck because it felt like it was originally written to be an eroge (suave and eternally selfless dude helps screwed up girls with their problems Key style) but I don't regret watching it either. I would have probably enjoyed it more if the direction was more focused. Shinbo does that weird ass ADHD Family Guy-tier direction throughout the whole series (like 10 scene changes per second, random shit constantly occupying and popping up on the screen) and it personally leaves me exhausted. However the female characters had me interested enough to keep on watching (I'll watch the other seasons too). 8 autism bucks out of 10.

I'm trying to get back into watching anime, so I've watched most of the first episodes of each new show.

I've also finished watching the Utawarerumono anime. I had read the VN long ago, but I recalled reading posts saying the anime was better and that it was one of the few cases the anime surpassed the VN. Boy, were they wrong. Don't even bother watching the anime. It's ok material. The VN is far superior in every aspect.

Back with my thoughts of Penguin Musume and Nogizaka Haruka.

I highly enjoyed Penguin Musume, which was a fun, silly series that requires a high powerlevel to fully appreciate. Penguin is the best girl by far and Etorofu-chan is a solid second. The plot was nonsensical for the most part and the subs (Chihiro) were bad but still managed to make me smile.

Rating: 7.5/10

I was expecting Nogizaka to be similar to Oreimo and while the show was good enough to finish the plot felt forced as fuck and many of the characters are extremely annoying. Haruka is a real qt 3.14 but calling her otaku is really stretching it.

Rating: 7/10

Spicy Wolf 2 is boring so far so I hope it picks up and Oretachi is all over the place I can't make sense of it at all. Alice-chan more than makes up for it though with her cute loli body :3

Haganai isn't a harem series, dude. The characters are so asexual when it comes to each other that it hurts. Maybe I can see it eventually turning into a love triangle series, but Meat/Yozora are too beta and autistic to actively pursue the main guy (which seems to be the main joke that the series is riding on). And so far even the protag doesn't see any of his friends as actual women, and quite literally so for one certain character.

If you want a fun and recent harem series proper, then I suggest Campione!

>>Yes, and "crossboarder" is a 2012 term.
>That's why it's been used at least since 2009. Right. I guess all the earlier occurences are a result of someone randomly hitting the keyboard, not just a different grammatical form of a long established term.

>>the term "cross" and the term "board" both date back thousands of years.
>And the letters c, r, o, s, b, a and d date even further back in time. This only "makes sense" under the same reasoning that makes you treat words as chains of letters, disregarding their semantical and ontological properties. The meaning of the term back in 2008 was already the same as it is now. "This is more or less indisputable."

In studies of nucleotide synthesis, Parnas and Ostern (1929) found that the formation of adenosine monophosphate (AMP) from inosine monophosphate required input of nitrogen from an organic amino group donor and not from ammonia (see Fig 22.34).
In 1930 Dorothy Needham was examining the effects of adding aspartate or glutamate to fresh extracts of skeletal muscle. She observed that the amount of amino acids declined rapidly after addition to the muscle extracts and this was accompanied by the formation of nitrogen-free products such as succinate. She expected the amino group nitrogen of the amino acids to emerge as ammonia, urea or uric acid but none of these products were found.
Needham’s "missing nitrogen" was later inadvertently explained by Alex Braunstein, who was testing glutamate as the potential organic amino group donor predicted by Parnas. In fact, this didn't work because aspartate is the real donor (look for details of IMP metabolism in Chapter 22 of the text). After adding glutamate to muscle extracts, Braunstein noticed the disappearance not only of the added glutamate but also of lactate that was already present in the muscle extract. Although the initial aim of his experiment was unsuccessful, he went on to carefully analyze the products and found alpha-ketoglutarate and alanine. He concluded that the amino group of the added glutamate was transferred to pyruvate, leading to formation of alanine. He predicted that pyruvate was the most likely acceptor for the amino groups because it could be generated from muscle lactate by the actions of lactate dehydrogenase. Based on these observations Braunstein concluded that the muscle extracts contained "aminoferases", later called aminotransferases or transaminases.

“Equilibration” of amino groups revealed with [15N]-labeled amino acids
Subsequent studies have shown that a large number of transaminases play vital roles in vivo. For example, injection of one amino acid isotopically labeled with “heavy” nitrogen [15N] leads to rapid appearance of the isotopic label in many other amino acids. Over the time intervals required for such studies, typically 1-2 hours, the concentrations of each of the amino acids in the blood do not significantly change and the results therefore demonstrate that the amino-groups are exchangeable in vivo. The amino groups of lysine and threonine are exceptional, exchanging extremely slowly over a 2-hour period.

ROLE OF PYRIDOXAL PHOSPHATE (PLP) IN TRANSAMINATION
Transamination occurs in two stages or half-reactions and the reaction is absolutely dependent on the presence of pyridoxal phosphate in the enzyme active site:

1. The amino group donated by the reacting amino acid is transferred to pyridoxal phosphate (PLP), forming pyridoxamine phosphate coupled with the release of the keto acid product.
2. In the second half reaction, the incoming keto acid accepts the amino group to form the amino acid product with regeneration of pyridoxal phosphate.

PYRIDOXAL PHOSPHATE IS DERIVED FROM PYRIDOXINE (VIT B6)
Pyridoxal phosphate (or pyridoxamine phosphate) is derived from the vitamin B6, which exists in several forms (vitamers) – pyridoxine, pyridoxal and pyridoxamine. The vitamers require one or both of two reactions to generate the active coenzyme. One hydroxyl substituent of the vitamin must be oxidized to the corresponding aldehyde and the second hydroxyl must be phosphorylated.

Pyridoxal phosphate and pyridoxamine phosphate can be interconverted in the presence of keto and amino acids even in the absence of enzymes:

pyridoxal phosphate + amino acid  pyridoxamine phosphate + keto acid

Furthermore, inactive preparations of apo-transaminase can be activated by the addition of either pyridoxal phosphate or pyridoxamine phosphate:

Apo-transaminase + pyridoxal phosphate   holo-transaminase

Much further work has been done to define the structure and functions of the large family of more than sixty transaminases. Structural studies have shown that PLP is held in the active site cleft of transaminases by several types of interactions:

ahe phosphate ester and ionizable hydroxyl of PLP provide ionic interactions
(b) The methyl substituent of PLP interacts hydrophobically
(c) The aldehyde of PLP forms a Schiff Base (aldimine) with the amino side-chain of a lysine residue at the enzyme active centre

>> No.10348119,249 [INTERNAL]

THE TRANSAMINASE REACTION MECHANISM
The steps of the proposed reaction mechanism (see Chapter 18) are as follows:
1. The in-coming amino acid substrate (amino donor) enters the active site and displaces the active-site lysyl residue to form a Schiff base with pyridoxal phosphate (by transimination).
2. The amino acid alpha carbon is de-protonated by a basic group of the enzyme active site, generating a carbanion intermediate.
3. The carbanion can be stabilized by electron delocalization through the conjugated PLP structure, accounting for resonance stabilization and reduction in activation energy.
4. "Reverse" electron rearrangement then allows the [H] to be added back; NOT to the original alpha carbon but instead to the PLP aldehyde carbon.
5. Hydroxylation of the alpha carbon of the amino acid is followed by generation of a keto group and therefore the keto acid that becomes the leaving group. The amino group remains attached to coenzyme, as pyridoxamine phosphate.
6. This represents the half-way point of the overall reaction. A different keto acid must now enter the active site and the sequence of steps is reversed to release a corresponding amino acid.

ADDITIONAL REACTIONS INVOLVING PYRIDOXAL PHOSPHATE
PLP is used in a variety of enzymes in addition to the transaminases, alternative reactions including:
Decarboxylation, as in the generation of bioactive amines
Racemization, for the interconversion of D- and L- amino acids
Removal of or reactions within the R group

The MATa1 gene encodes a transcriptional repressor
that is an important modulator of sex-specific
gene expression in Saccharomyces cerevisiae.
MATa1 contains two small introns, both of which
need to be accurately excised for proper expression
of a functional MATa1 product and to avoid production
of aberrant forms of the repressor. Here, we
show that unspliced and partially spliced forms of
the MATa1 mRNA are degraded by the nuclear
exonuclease Rat1p, the nuclear exosome and by
the nuclear RNase III endonuclease Rnt1p to
prevent undesired expression of non-functional a1
proteins. In addition, we show that mis-spliced
forms of MATa1 in which the splicing machinery
has skipped exon2 and generated exon1–exon3
products are degraded by the nuclear 50–30 exonuclease
Rat1p and by the nuclear exosome. This
function for Rat1p and the nuclear exosome in the
degradation of exon-skipped products is also
observed for three other genes that contain two
introns (DYN2, SUS1, YOS1), identifying a novel
nuclear quality control pathway for aberrantly
spliced RNAs that have skipped exons.

Nucleosomes are surprisingly dynamic structures
in vivo, showing transcription-independent exchange
of histones H2A-H2B genome-wide and exchange of
H3-H4 mainly within the promoters of transcribed
genes. In addition, nucleosomes are disrupted in
front of and reassembled behind the elongating
RNA polymerase. Here we show that inactivation of
histone chaperone Spt16 in yeast results in rapid
loss of H2B and H3 from transcribed genes but
also from inactive genes. In all cases, histone loss is
blocked by a transcription inhibitor, indicating a
transcription-dependent event. Thus, nucleosomes
are efficiently evicted by the polymerase but do
not reform in the absence of Spt16. Yet exchange
of nucleosomal H2B with free histones occurs
normally, and, unexpectedly, incorporation of new
H3 increases at all loci tested. This points to Spt16
restoring normal nucleosome structure by redepositing
the displaced H3-H4 histones, thereby preventing
incorporation of new histones and perhaps changes
in histone modification patterns associated with
ongoing transcription.

How the directionality of vesicle traffic is achieved remains an important unanswered question in cell biology. The
Sec23p/Sec24p coat complex sorts the fusion machinery (SNAREs) into vesicles as they bud from the endoplasmic
reticulum (ER). Vesicle tethering to the Golgi begins when the tethering factor TRAPPI binds to Sec23p. Where the
coat is released and how this event relates to membrane fusion is unknown. Here we use a yeast transport assay to
demonstrate that an ER-derived vesicle retains its coat until it reaches the Golgi. A Golgi-associated kinase, Hrr25p
(CK1d orthologue), then phosphorylates the Sec23p/Sec24p complex. Coat phosphorylation and dephosphorylation are
needed for vesicle fusion and budding, respectively. Additionally, we show that Sec23p interacts in a sequential manner
with different binding partners, including TRAPPI and Hrr25p, to ensure the directionality of ER–Golgi traffic and
prevent the back-fusion of a COPII vesicle with the ER. These events are conserved in mammalian cells.

Phosphorylation of the RNA polymerase (Pol) II C-terminal domain (CTD) repeats (1-YSPTSPS-7) is coupled to transcription and
may act as a ‘code’ that controls mRNA synthesis and processing. To examine the code in budding yeast, we mapped genome-wide
CTD Ser2, Ser5 and Ser7 phosphorylations and the CTD-associated termination factors Nrd1 and Pcf11. Phospho-CTD dynamics
are not scaled to gene length and are gene-specific, with highest Ser5 and Ser7 phosphorylation at the 5􀁠 ends of well-expressed
genes with nucleosome-occupied promoters. The CTD kinases Kin28 and Ctk1 markedly affect Pol II distribution in a gene-specific
way. The code is therefore written differently on different genes, probably under the control of promoters. Ser7 phosphorylation
is enriched on introns and at sites of Nrd1 accumulation, suggesting links to splicing and Nrd1 recruitment. Nrd1 and Pcf11
frequently colocalize, suggesting functional overlap. Unexpectedly, Pcf11 is enriched at centromeres and Pol III–transcribed genes.

AMONGST the most striking remains of the earliest dynasties at Abydos are those
groups of graves called by their excavator the Tombs of the Courtiers.' They lie on the
low gravel plateau behind the site of the early town of Abydos and the temple of
Khentiamentiu, and consist of three groups of small brick-lined graves, for the most
part constructed in continuous trenches, and so arranged as to enclose large rectangular
areas, the largest of which measures some 75 metres by 123 metres, and contains about
280 graves. On the analogy of similar burials, subsidiary to large mastabas, in particular
around Saqqara tomb 3504,2 each of these small graves would originally have been
marked on the surface by a low rectangular brick superstructure with a slightly vaulted
roof, and perhaps with one or two small niches. From the contents of these graves,
two of the groups were assigned to the reigns of Djer3 and Djet,4 and the thirdlargely
on the basis of a pot-mark-to the period or reign of Merneith.5 The function
of these rows of small graves has long been something of a puzzle for, while they seem
very similar to the subsidiary burials which commonly surrounded royal and other
large tombs of the First Dynasty, a search by Petrie in the area inside these rows
failed to show any comparable structure to which these graves could have been subsidiary.
There has, however, been no lack of explanations. According to Reisner,6
following a suggestion by Petrie,7 each group originally surrounded an enormous brick
mastaba of the 'palace-facade' type similar to the royal tomb at Naqada and without
substructure. They would have served as Valley Temples to the royal tombs on the
Umm el-Qaa^b. This theory was subsequently dismissed as being incompatible with
what is known of later Valley Temples.8 Helck later concluded that inside each rectangle
was constructed an imitation palace built probably of matting and intended to
house a statue of the deceased king, and saw this as a development of burial customs
attributed to the earlier inhabitants of Upper Egypt of supposed nomadic origin.

The Old and Middle Kingdoms together represent an important unitary
phase in Egypt's political and cultural development. The Early Dynastic
Period had seen the creation and consolidation of a type of government
and court culture which, with the Third Dynasty, now reached levels
of scale and competence marking the beginning of the plateau of
achievement for ancient Egypt. After five centuries and following the
end of the Sixth Dynasty (c. 2181 BC) the system appears to have
faltered, and there seems to have ensued a century and a half of
provincial assertion and civil war, the First Intermediate Period. But
the re-establishment of powerful central government which followed,
c. 2040 BC, seems to have been, with certain changes of nuance, the
re-establishment of the patterns of the Old Kingdom. There is thus
much to be said for treating certain important aspects of the Old and
Middle Kingdoms together.

Divine kingship is the most striking feature of Egypt in these periods.
In the form of great religious complexes centred on the pyramid tombs
its cult was given monumental expression of a grandeur unsurpassed
anywhere in the ancient Near East. Yet despite its all-pervading
influence in Egyptian civilization it is not easy to present a coherent
account of its doctrines, especially one which avoids mixing material
from widely separated periods. One good reason for this is the Egyptian
mode of communication, presenting doctrine not in the form of
cogently argued treatises intended to persuade, but as series of concisely
worded assertions which to us often take on a deeply cryptic appearance.
The basic assertions are that the king is the holder of an office which
is divine, he is 'the good god'; that he is a particular incarnation of
Horus, an ancient sky and falcon god who became closely linked with
the sun cult of Ra; that he is a son of Ra, the sun god, something

incorporated into royal titulary from the Fourth Dynasty onwards. In
the latter part of the Old Kingdom the deceased king became identified
with Osiris, a god of the dead standing in a special relationship to the
kingship.
For the periods under consideration three important texts, or groups
of texts, deal with divine kingship. One is the Memphite Theology,
known from an eighth-century BC copy of a document composed much
earlier, possibly in the Old Kingdom or even before, although this is
a disputed matter. It attempts to explain the geographical duality of
Egyptian kingship, the positions of the gods Horus and Seth, and the
supremacy of the capital city of Memphis and ultimately of its creator
god, Ptah. Horus is presented as the first king of Upper and Lower
Egypt, acquiring this position, having been earlier only the king of
Lower Egypt, after the god Geb had given him also the kingship of
Upper Egypt, hitherto held by Seth. The mythically aetiological element
is so manifest that it is pointless to search for strictly historical features,
particularly since the picture it suggests is at variance with the
archaeological record. The second is the Ramesseum Dramatic Papyrus,
dating to the reign of Senusret I (c. 1971 BC). It contains forty-six scenes,
illustrated by thirty-one drawings, and includes instructions for the
performance of ritual acts. The rituals, accompanied by notes on their
mystic significance, seem intended for the king's accession or for his
jubilee ceremony, and we may presume, therefore, that with this text we
are confronted with ideas at the very heart of the Egyptians' concept
of kingship. We find that it is concerned primarily with the king's
relationship to Horus, Osiris, and Seth, to the very situation for which
the Memphite Theology offers its 'historical' explanation. The Pyramid
Texts, inscribed in the subterranean parts of the pyramids of kings from
Unas to Pepy II, and Aba of the Eighth Dynasty, and of three late Sixth
Dynasty queens form the third main source. Although their language
is seemingly an archaic one, those who edited the texts for a particular
pyramid would seem to have had sufficient working knowledge of it
to adapt them to changing revelations, and even perhaps to compose.
The increase and change in nature of allusions to Osiris and to Seth
is one demonstration that they represent a living tradition. Their
purpose is to assert the king's supremacy as a god, after rebirth, in a
many-sided afterlife. Although the Horus-Osiri

Oh look, you're using your homo smilies again. Did the /jp/ gay slayers not catch up to you yet?

I guess I was a bit too generous with that head-start I gave you.

>> No.10348119,277 [INTERNAL]

>>10348119,276
You really enjoy roleplaying don't you Trev-Trev. Okay :3

They caught up to me but they let me go, it looks like they only go after gay people. They apologized for the inconvenience, and bought me a bouquet of 15 yellow roses. Overall really nice people, the bouquet was definitely really nice of them.

>> No.10348119,278 [INTERNAL]

>>10348119,277
Funny because they haven't called me on my walkie-talkie yet.

You can't bullshit a bullshitter™.

>>10348119,278
I told them one yellow rose would have also been appropriate, but I guess with roses you really need to have a bouquet.

I have them displayed on my dining room table right now, in a nice green vase. Its a really nice vase too, the glass looks really great on a nice summer day.

That post really isn't helping your I'm-not-gay claim.

I await seeing your name taken off the APB so I can post on warosu in peace again.

No I'm quite certain that it does. Anyways, I'm just such a nice person that I figured I'd humor your delusions. You can thank me if you wish, I'll be here to receive your words of gratitude Trev-Trev.

Oh and you might want to refresh, you'll see that I was removed and a letter of apology was issued. Like I said, they were proper gentlemen.

>> No.10348119,283 [INTERNAL]

>>10348119,282
Funny because I actually did refresh the APB and it says your bounty was doubled for being much gayer than originally described. They needed to retreat and put on more protective layers so they weren't infected by the record-breaking levels of gay being radiated off of your incredibly homosexual person.

Did you seriously think I wouldn't check and just take your word for it?

You're not dealing with some half-assed amateur here. There's a reason why other people do this work for me.

>> No.10348119,284 [INTERNAL]

>>10348119,283
You... you really enjoy this kind of thing... don't you? Alright, if I must, only because I think its in the best interest of your mental health.

I fold, I must admit that I really enjoy Yuri, but none of that overly sexual stuff. I just really enjoy the light-hearted romances with lots of cuddling and such. I mean I guess I enjoy the sexy stuff too, but that has its own time and place, and on a much more rare occasion.

The team notified me while they were here that they thought it was extremely cute, and in their embarrassment, decided to take their leave. They increased the bounty to make it seem like I would be a tough case to deal with, thereby persuading people to stay away for their own safety. Quite nice of them if you think about it, I suppose they'll be "retreating" for quite a long duration.

>> No.10348119,285 [INTERNAL]

>>10348119,284
How is a dude looking at yuri gay? Are you fucking retarded?

Hell, why am I even asking? You obviously are if you fell for such an obvious tactic. If that actually did happen it was so that they could hire a third party at the original bounty reward (obviously before it was doubled, normally I wouldn't have to point this out but clearly you're pretty fucking stupid and need to explained) to carry out the contract and split the doubled bounty, thereby receiving the same reward without having to damage any of their gay-handling equipment and pay for repairs which I can only imagine would be incredibly costly if you're even half as gay as it seems.

I hope you enjoy being stuck inside a vacuum tube for eternity. Say hello to Casper for me if you see him in there.

>> No.10348119,286 [INTERNAL]

>>10348119,285
Well for one I'm not a boy, I don't know how painfully obvious I have to make this, so theres that out of the way. I guess I shouldn't have expected anything out of someone as completely dense as you.

I guess we'll just have to wait and see, they seemed quite sincere when they apologized. Maybe you just have difficulty reading people? It would definitely fit the bill of someone with such an extreme case of autism. Yeah, I'm sure thats how it is, how cute of you to be so naive!

I guess its a good thing I won't have to stick around in isolation: I like to have a mojito here and there, you know? Casper was a really heartwarming film, I haven't seen it in since it came out, but I remember it fondly.

I finally decided to marathon chuu2 and is this show ever depressing. Why are all the characters so hard on the middle school girl? Its like they all went to great lengths to absolutely destroy her emotionally. What a performance by her voice actress though, almost found myself crying after a few scenes.

I just finished the last episode, and I have to say the show was really good all around. It started out entertaining and then transitioned to a really depressing drama. I'll have to say that by the end my heart was in my throat; it really is a privilege to be able to say goodbye, in any form.
