I opened it in PSP, masked the layer according to the luminance, then inverted the mask. I'm hosting this file on a web server I'm running on my computer.

I copied my background element(s) to .reply, .replyhl to insert it and modified the colour aspect to return to a good approximation of the original reply box colour. There is likely a better method somewhere.

I'm making use of all 8 bits of PNG alpha in my image, to provide a range of transparency. I based the alpha values off the inverse of the luminance of the relevant pixel. Since I use PSP, this involved defining a mask for a layer.

Then you use the same background CSS for the "reply" and "replyhl" classes to get the image to display when a reply box overlays the image in the background.
