I always thought it was about Gregor lusting after, & womanising his sister.

At the start he's being like a grandparent that spoils a child by wanting to send her to music school just so he can dick her against the wiser & more maternal instincts of his parents (he should share these as a brother but doesn't because he's such a sex obsessed freak). Then due to the repression of these feelings he becomes like a "bug/vermin" to the rest of the family (just a Kafkaesque metaphor of course) & generally just becomes a moody & difficult prick, always needing extra help to deal with his issue. Even the maid gets fed up with his attitude towards women. Then he allows his sister to take the blame for his incestuous actions before finally making a move towards her in the whole violin scene.

At the end there's the triumph of the "family instinct", or w/e you want to call it, over the cancer/unnatural/rebellious attitude of Gregors incestous actions & this is celebrated by Gregor's sister's maturation into a full grown woman, complete with a job as any man would have.
