I don't know if this is some kind of meme as I'm not a frequent /lit/ user, but no, that is not correct usage.

A semicolon is only to be used in place of a comma with a coordinating conjunction. You use it to separate two independent clauses. The way you used it is incorrect because "so" is a coordinating conjunction. To rephrase it, it would be, "I answered all of the questions on it; could I make it up?" However, it would be a bad idea to use a semicolon in this place because it would be totally unnecessary. Reserve semicolons for when the two sentences are closely linked in idea or are completely identical grammatically. In this case, the ideas are linked, but in a causal way and ending in a question, so the semicolon is inappropriate. It would be better to simply write out, "I answered all of the questions on it. Could I make it up?"
The only other time you use a semicolon is to separate items in a list that already contain commas in order to avoid confusion.
